Monoclonal antibody production
from DNA sequence file
From sequence file to monoclonal antibody (approx. duration: 7 months)
Antibody production starting from a DNA sequence file
GENEART offers the production of antibodies by genetic immunization starting with nothing more than a DNA sequence file.
After receiving the sequence information from a customer, e.g. by e-mail, the gene will be produced synthetically using GENEART’s integrated in-house gene synthesis process which includes gene optimization and design, oligonucleotide synthesis and gene assembly. The use of expression-optimized genes for immunization often increases the success of antibody production. Sometimes the complete gene does not have to be produced for immunization. GENEART is able to define specific regions of your target antigen that are well suited to inducing antibody response in the host after genetic immunization.
Immunization with verified expression vector from customer possible.
We deliver up to three subcloned hybridoma lines and all positive parental hybridoma with 5 ml supernatant each (if required).
Service includes:
- production of synthetic gene optimized for expression in the immunized animal
- expression control of recloned DNA construct
- subcloning of customer gene into optimized immunization vector
- genetic immunization of animals (2 rabbits, 5 rats, 5 mice)
- all animal care and handling
- titer determination
- hybridoma generation and screening
- subcloning of selected hybridoma lines
- milestone system for customers (see below)
Delivery times
The average delivery time for the hybridoma lines depends on the immune response to the antigen (number of immunizations) and the amount of subcloning. It usually takes 6-7 months. Animals are immunized until a satisfactory titer is detected.
Hybridoma cells are screened for antibody production and the best candidates are subcloned.
Milestone system for customers
If the project fails to reach one of the following milestones, it can be stopped and customers then only pay for the work that has actually been carried out.
Milestone 1: Expression control of subcloned gene
Milestone 2: Immunization of 5 mice or rats
Milestone 3: Fusion and selection of clones
We provide no guarantee of successful project completion.
Prices
Prices differ depending on the number of antibodies ordered. If antibodies against more targets are ordered at the same time and can be processed in parallel, prices are lower.
| Number of targets In parallel production |
Price per target |
| 1 | €12,500 |
| 2 | €10,000 |
| 3 | €8,500 |
Synthetic gene costs depend on the gene length.
Terms of payment
20% when order placed
20% when immunization starts
60% when fusion starts
