Polyclonal antisera production

from DNA sequence file

Service 1: From sequence file to antiserum (approx. duration: 4 months)

Antibody production starting from a DNA sequence file

If you have nothing but the sequence file of your protein available, GENEART can synthesize a gene that is optimized for immunization. You simply provide a data file or accession number regarding the gene sequence of target protein.

Service includes:

  • increased control for immunization success through analysis of target protein using sophisticated software and well-trained scientists
  • production of synthetic gene optimized for expression in the immunized animal
  • expression control of recloned DNA construct
  • subcloning of customer gene into optimized immunization vector
  • preparation of plasmid DNA for immunization
  • preparation of ammunition for gene gun application
  • immunization of animals (2 rabbits, 5 rats, 5 mice)
  • animal care
  • pre-bleeding and final bleeding
  • titer determination on transfected eucaryotic cells using FACS analysis
  • milestone system, i.e. if expression of recloned immunization construct is negative, the customer only pays 50%

Service 2: Optimized antisera production (approx. duration: 3 months)

Customer provides 1-5 µg of plasmid DNA with the relevant gene cloned into any plasmid vector. A few micrograms of the vector in an eppendorf tube or simply on a small piece of filter paper will be sufficient.

Service includes:

  • increased control for immunization success through analysis of target protein using sophisticated software and well-trained scientists
  • expression control of recloned DNA construct
  • subcloning of customer gene into optimized immunization vector
  • preparation of plasmid DNA for immunization
  • preparation of ammunition for gene gun application

  • titer determination on transfected eucaryotic cells using FACS analysis
  • milestone system, i.e. if expression of recloned immunization construct is negative, the customer only pays 50%

Service 3: Antisera production with expression construct provided by customer (approx. duration: 3 months)

Customer provides 1 mg of column-purified plasmid DNA with the relevant gene cloned into an expression vector that has been tested for expression of the relevant protein in mammalian cells. Service only recommended for secreted proteins.

Service includes:

  • preparation of ammunition for gene gun application
  • immunization of animals (2 rabbits, 5 rats, 5 mice)
  • animal care
  • pre-bleeding and final bleeding

Delivery times

Average delivery time for polyclonal antibodies: 3 months depending on number of immunizations.

Amount of antisera

We provide at least 70 ml of antiserum per rabbit, 0.2 ml per mouse and 2-4 ml per rat.

Milestone system for customers, valid for polyclonal service 1 and polyclonal service 2 only

Customer only pays for services provided.
If expression is negative, the project can be stopped after the unsuccessful milestone (milestone 1) to limit the cost to our customers.

Milestone 1: Expression control of subcloned gene
Milestone 2: Immunization of 2 rabbits or 5 rats or 5 mice

We provide no guarantee of successful project completion.

Terms of payment:

For service including titer detection:
50% when order placed
50% on commencement of immunization

For service without titer detection:
100% when order placed

GENEART’s antibody synthesis service is based on a service fee only model and makes no claim on our customers’ intellectual property.
All IP remains with the customer.
