In cases where there are very high GC contents, direct/indirect repeats or long polypurine/pyrimidine runs, we have to use specially modified and purified oligonucleotides and special gene synthesis methods. Moreover, complex operon structures and very long natural sequences require highly sophisticated assembly strategies.
